CJC-1295 is a synthetic analogue of growth hormone-releasing hormone (GHRH), consisting of 30 amino acids. It has been shown to significantly boost growth hormone secretion and IGF-1 levels without adversely impacting the natural pulsatility of GH release.
Typically, CJC-1295 is paired with Ipamorelin, which offers greater specificity as a GHRH. This peptide raises growth hormone levels similarly, but does not induce appetite stimulation or increase cortisol, acetylcholine, prolactin, or aldosterone as some other peptides do. Ipamorelin is well-tolerated and synergizes well with CJC-1295.

Two Forms of CJC-1295:GHRH is produced in the hypothalamus, and its pulsatile release stimulates the pituitary gland to release growth hormone. GHRH has a very short half-life of just a few minutes.
The active segment of GHRH comprises the first 29 amino acids and is available as a synthetic peptide known as Sermorelin. Sermorelin has been modified to extend its half-life to 30 minutes, resulting in CJC-1295. Additionally, CJC-1295 was further altered by incorporating a Drug Affinity Complex (DAC), which binds to the blood protein albumin, extending its half-life to 8 days. This version is referred to as CJC-1295 + DAC. CJC-1295 can also be created in a non-DAC form, resembling a more natural GH spike occurring each night.
Due to the prolonged half-life from DAC’s interaction with albumin, injections are needed only once or twice a week. However, this long half-life and relatively stable blood levels may lead to a continuous stimulus for GH release from the pituitary via the GHRH receptor, which is not physiological. This can reduce the amplitude of GH pulses, potentially resulting in decreased stimulation of GH tissues.
Safety:It is advisable to take 'hormone holidays' of three months every three to six months when using a long-acting CJC molecule to allow the pituitary gland to recover. During these breaks, Sermorelin can be used instead of CJC-1295 + DAC.
These 'hormone holidays' may also help reduce the risk of developing GH resistance. Such resistance could occur if antibodies form that bind to and deactivate GH, or if there is a down-regulation of GH receptors in tissues. However, these concerns are theoretical, as there have been no long-term studies conducted to investigate these issues.
