CJC-1295 is a synthetic analogue of growth hormone-releasing hormone (GHRH) consisting of 30 amino acids. It has proven to be very effective in stimulating the secretion of growth hormone and IGF-1, while maintaining the natural pulsatility of GH release.
CJC-1295 is frequently paired with Ipamorelin because of its greater specificity as a GHRH. This peptide also promotes growth hormone secretion but does so without the appetite-stimulating effects and increases in cortisol, acetylcholine, prolactin, and aldosterone that can occur with other peptides in its class. It is well-tolerated and works effectively when combined with Ipamorelin.

Two Forms of CJC-1295:GHRH is generated in the hypothalamus and its pulsatile release triggers a corresponding pulsatile release of GH from the pituitary gland. GHRH has a very brief half-life of just a few minutes.
The active section of GHRH consists of the first 29 amino acids, which can be manufactured as a peptide known as Sermorelin. Sermorelin was modified to extend its half-life to 30 minutes, resulting in CJC-1295. Further modification of CJC-1295 by adding DAC (Drug Affinity Complex) allows it to bind to the blood protein albumin, extending its half-life to 8 days, known as CJC-1295 + DAC. There is also a non-DAC version of CJC-1295 that simulates a more natural GH spike each night.
The extended half-life with DAC means that injections are needed only once or twice a week. However, this long half-life and relatively steady blood levels can provide a continuous stimulus for GH release through the GHRH receptor, which may not mimic natural physiological conditions. This could potentially reduce the amplitude of the GH pulse and lessen its tissue stimulating effects.
Safety Considerations:It is advisable to take "hormone holidays" for three months every three to six months when using a long-acting CJC molecule, to allow the pituitary gland to "recover." During these breaks, Sermorelin can be used as an alternative to CJC-1295 + DAC.
These hormone holidays may also help minimize the risk of developing GH resistance, which could arise from antibodies that inactivate GH or a decrease in the number of GH receptors through down-regulation. These concerns remain theoretical due to the lack of long-term studies.
Potential side effects of CJC-1295 may include reactions at the injection site (such as irritation, redness, swelling, pain, and itching), headaches, diarrhea, vasodilation (flushing, warmth, temporary drops in blood pressure), nausea, and abdominal discomfort.
