Sunrise’s Malegra DXT combines two active ingredients—Sildenafil and Duloxetine. Sildenafil is primarily used to manage erectile dysfunction, while Duloxetine addresses premature ejaculation, together enhancing an individual's sexual experience. Although Malegra DXT is typically a prescription medication, it can be purchased from online pharmacies without a prescription. However, due to the potential for serious side effects when combined with certain medications, it is crucial to consult a doctor before use. Malegra DXT is available solely in oral capsule form.

During sexual arousal, nitric oxide is released from the corpus cavernosum, which is essential for regulating penile erection in men. Nitric oxide activates an enzyme called guanylate cyclase, leading to an increase in cGMP (cyclic guanosine monophosphate) levels. This process promotes relaxation of the smooth muscle within the corpus cavernosum, allowing more blood to flow through the penile artery. Sildenafil enhances the effect of nitric oxide by inhibiting the action of PDE5 (Phosphodiesterase type 5), which would otherwise break down cGMP in the penis. It’s important to note that Sildenafil requires external sexual stimulation to be effective, as it activates when arousal triggers the release of nitric oxide. Administered orally, Sildenafil is quickly absorbed, typically within one hour of ingestion, through hepatic metabolism primarily involving cytochrome 3A4 and P450 3A4, leading to the formation of an active metabolite.

Duloxetine, a thiophene derivative, functions as a selective serotonin reuptake inhibitor, which helps reduce stress on the brain during sexual activities. This effect slows down nerve sensation in the glans penis, thereby increasing the duration of intercourse and preventing premature ejaculation.

MALEGRA-DXT serves a dual purpose in treating sexual dysfunction, containing 100 mg of Sildenafil and 30 mg of Duloxetine.

First, Sildenafil is absorbed in the liver within 30 minutes, leading to increased blood flow in the corpus cavernosum, allowing for an erection typically achieved within one hour of taking the medication, lasting up to 4 to 5 hours.

Second, Duloxetine acts on the nerve centers to alleviate anxiety during sexual intercourse, which aids in preventing premature ejaculation.

MALEGRA-DXT comes in two dosages: 75 mg and 130 mg, with physicians typically recommending the 130 mg dose for better efficacy. It is advised to take one tablet one hour before planned sexual activity, and doctors caution against using it more than once a day.

While MALEGRA-DXT is generally considered safe and effective, it may result in minimal or no side effects in most individuals. Side effects occur mainly due to the dilation of blood vessels and can include nausea, insomnia, flushing, muscle pain, dyspepsia, dizziness, vomiting, loose stools, epistaxis, and erythema.

MALEGRA-DXT may lower blood pressure if used in conjunction with alpha blockers. Additionally, its interaction with erythromycin or cimetidine can prolong penile erection.
