Sermorelin is a synthetic form of growth hormone-releasing hormone (GHRH), which is the natural hormone that stimulates the production of growth hormone in the body. Also referred to as growth hormone releasing factor and GRF 1-29, it is classified as a growth hormone secretagogue, a compound that encourages the pituitary gland to release more growth hormone.

Mechanism of ActionSimilar to the body’s natural GHRH, Sermorelin binds to the growth hormone-releasing hormone receptor (GHRHr) located in the anterior pituitary gland. This action stimulates the production of growth hormone and promotes an increase in endogenous IGF-1 levels. Due to its resemblance in function to GHRH, the positive impact of Sermorelin on growth hormone release is regulated by a negative feedback mechanism driven by somatostatin, also known as growth hormone-inhibiting hormone (GHIH). This process prevents excessively high levels of growth hormone and mimics the natural fluctuations typically seen in growth hormone concentrations.
The natural mechanism of action of Sermorelin helps maintain the hypothalamic-pituitary-somatotropic axis, thus preventing the suppression of hormone production that often occurs with direct human growth hormone (hGH) replacement. Additionally, Sermorelin has been shown to enhance sleep quality by promoting longer periods of restorative slow-wave sleep, increasing sleep-related growth hormone secretion while lowering cortisol levels, which is a catabolic stress hormone.

Although Sermorelin has a relatively short half-life of about 20 minutes, it has been shown to trigger a significant increase in growth hormone release up to two hours after administration. Protocols utilizing three daily doses of as little as 100 mcg (0.10 mg) each have led to notable increases in growth hormone and IGF-1 levels.
