CJC-1295 is a synthetic analogue of growth hormone-releasing hormone (GHRH) composed of 30 amino acids. Research indicates that it significantly enhances the secretion of growth hormone and IGF-1 without disrupting the natural pulsatile release of GH.

It is commonly paired with Ipamorelin, which offers increased specificity as a GHRH. This combination results in similar enhancements in growth hormone secretion while avoiding the appetite stimulation and elevated levels of cortisol, acetylcholine, prolactin, and aldosterone commonly associated with other peptides in its category. Users generally find this peptide very tolerable, making it an ideal partner for Ipamorelin.

GHRH is naturally produced in the hypothalamus and stimulates the pituitary gland to release GH in a pulsatile manner. GHRH has a very short half-life of merely a few minutes—meaning it is quickly eliminated from the bloodstream, leading to brief effects.

Types of CJC-1295:

The active portion of GHRH is composed of the first 29 amino acids and is commercially available as the peptide Sermorelin. Sermorelin was further modified to extend its half-life to 30 minutes, resulting in CJC-1295. This, in turn, was enhanced by the addition of a Drug Affinity Complex (DAC) that attaches to the blood protein albumin, prolonging its half-life to about 8 days, referred to as CJC-1295 + DAC. CJC-1295 can also be available in a non-DAC form that mimics a physiological GH spike occurring nightly.

The extended half-life due to DAC means that injections can be limited to once or twice weekly. However, this sustained presence in the bloodstream can create a constant stimulation of GH release via the GHRH receptor, which may not reflect natural physiological patterns. It can reduce the amplitude of GH pulses, potentially leading to less stimulation of GH-responsive tissues.

When using a long-acting CJC molecule, it is advisable to take "hormone holidays" every three to six months, lasting three months each, to allow the pituitary gland to recuperate. During these breaks, Sermorelin is recommended as a substitute for CJC-1295 + DAC.

Safety Considerations:

These 'hormone holidays' may also help lessen the risk of developing GH resistance, which could occur due to antibodies that neutralize GH or a reduced number of GH receptors on tissues (down-regulation). These concerns remain theoretical, as there haven't been extensive long-term studies conducted to address them.

Possible side effects of CJC-1295 include reactions at the injection site (such as irritation, redness, swelling, pain, and itching), headaches, diarrhea, vasodilation (including flushing, warmth, and transient low blood pressure), nausea, and abdominal discomfort.
