CJC-1295 is a synthetic analogue of growth hormone releasing hormone (GHRH), consisting of 30 amino acids. It is known for its efficacy in boosting growth hormone and IGF-1 levels without disrupting the natural pulsatility of GH secretion.

This peptide is frequently paired with Ipamorelin to take advantage of its greater specificity as GHRH. While Ipamorelin similarly promotes growth hormone release, it does so without the appetite increase and elevated levels of cortisol, acetylcholine, prolactin, and aldosterone associated with other peptides in its category. CJC-1295 is well-tolerated and is an excellent match when used alongside Ipamorelin.

    Types of CJC-1295:

    GHRH is synthesized in the hypothalamus, and its pulsatile release stimulates GH release from the pituitary gland. GHRH has a very short half-life of a few minutes, meaning it is quickly cleared from the bloodstream.

    The active portion of GHRH consists of the first 29 amino acids and is available as the manufactured peptide Sermorelin. This peptide has been modified to extend its half-life to around 30 minutes, resulting in CJC-1295. Further modification with DAC (Drug Affinity Complex) enables it to bind to albumin, lengthening the half-life to about 8 days, thus creating CJC-1295 + DAC. Additionally, CJC-1295 can be made without DAC to more closely replicate natural GH spikes at night.

    The extended half-life due to DAC means that injections are only needed once or twice a week. However, the continuous levels in the blood provide a constant stimulus for GH release from the pituitary via the GHRH receptor, which is not a typical physiological response. This could lead to a reduced amplitude of GH pulses and subsequent diminished GH tissue stimulation.

    Safety Considerations:

    For those utilizing a long-acting CJC compound, it is advisable to take "hormone holidays" of about three months every three to six months, allowing the pituitary to recover. During these breaks, Sermorelin can be substituted for CJC-1295 + DAC.

    "Hormone holidays" may also help avert the development of GH resistance, which can arise from the formation of antibodies that deactivate GH or due to a reduction in GH receptors in tissues (down-regulation). However, these are theoretical concerns, as there have been no long-term studies to definitively address them.

    Possible side effects of CJC-1295 may include reactions at the injection site (irritation, erythema, swelling, pain, itching), headaches, diarrhea, flushing, warmth, transient low blood pressure, nausea, and abdominal discomfort.
